Doxorubicin HCl is a chemotherapy drug that works by inhibiting the enzyme topoisomerase II, which is involved in DNA unwinding and repair. This drug is used to treat a wide variety of cancers, including solid tumors and hematologic malignancies, by killing cancer cells and preventing their ability to reproduce. Administered intravenously, doxorubicin is distributed quickly throughout the body and metabolized in the liver. It has a half-life of 20-48 hours and is excreted via bile and urine. Doxorubicin intercalates between DNA base pairs and inhibits the enzyme topoisomerase II, preventing DNA replication and transcription. This leads to the death of rapidly dividing cancer cells. Doxorubicin remains one of the most potent chemotherapeutic agents, widely used across multiple cancer types. However, its use is often limited by its dose-dependent cardiotoxicity, prompting ongoing research into drug delivery systems that can improve therapeutic outcomes and reduce adverse effects.
